Critical Swelling of Phospholipid Bilayers

چکیده
We reexamine a critical phenomenon of phospholipids in lamellar phases. The question is, Is the anomalous divergence in the repeat spacing near the main transition the result of a divergence of the water layer or of the lipid bilayer? X-ray diffraction of DLPC lamellae was measured in partially dehydrated conditions. Its critical behavior is much more pronounced than the previously studied DMPC. The bilayer thickness was calculated from dehydrated conditions and then extrapolated to full hydration. The results show that the anomalous divergence is primarily due to the water layer expansion. Anomalous swelling in phospholipid bilayers is not coupled to the formation of a ripple phase.
Aligned stacks of monomethyl and dimethyl dimyristoyl phosphatidylethanolamine (DMPE) lipid bilayers, like the much studied dimyristoyl PC (DMPC) bilayers, swell anomalously in a critical fashion as the temperature is decreased within the fluid phase towards the main transition temperature, T(M). Unlike DMPC bilayers, both monomethyl and dimethyl DMPE undergo transitions into a gel phase rather...

The Hofmeister series illustrates how salts produce a wide range of effects in biological systems, which are not exclusively explained by ion charge. In lipid membranes, charged ions have been shown to bind to lipids and either hydrate or dehydrate lipid head groups, and also to swell the water layer in multi-lamellar systems. Typically, Hofmeister phenomena are explained by the interaction of ...
متن کاملHydration process of multi-stacked phospholipid bilayers to form giant vesicles
– The hydration process of multi-stacked phospholipid bilayers to form giant vesicles was investigated by time-resolved small angle X-ray scattering. The hydration of lipids in the liquid-crystalline phase was found to proceed in two stages. In the early stage, until about 100 s after hydration, lipid bilayers on a solid substrate swell about 12 Å and reach a quasi-stable state. In the late sta...

Using neutron diffraction and a specially constructed high pressure cell suitable for aligned multibilayer systems, we have studied, as a function of pressure, the much observed anomalous swelling regime in dimyristoyl- and dilauroyl-phosphatidylcholine bilayers, DMPC and DLPC, respectively. We have also reanalyzed data from a number of previously published experiments and have arrived at the f...
متن کاملAtomic force microscopy study of thick lamellar stacks of phospholipid bilayers.
We report an atomic force microscopy (AFM) study on thick multilamellar stacks of approximately 10 microm thickness (about 1500 stacked membranes) of 1,2-dimyristoyl-sn-glycero-3-phoshatidylcholine deposited on silicon wafers. These thick stacks could be stabilized for measurements under excess water or solution. From the force curves we determine the compressional modulus B and the rupture for...
